Pharmacology
Quinethazone is a long-acting, quinazolinesulfonamide derivative with thiazide-like diuretic activity. Quinethazone acts in a similar manner to that of other typical thiazide diuretics and is no longer commercially available in the United States.
How it is made
Cohen et al, J Am Chem Soc 82, 2731 (1960), Cohen, Vaughan, Jr, US patent 2,976,289 (1961 to Am Cyanamid).
4'-Chloro-o-acetotoluidide is subjected to chlorosulfonation & subsequent amination to form 2-amino-4-chloro-5-sulfamoylbenzamide. Refluxing with an acidulated alcoholic solution of the diethylacetal of propionaldehyde effects the required condensation cyclization to yield quinethazone.
