Boronic acids and esters

Chemists use these reagents to join aromatic and heteroaromatic rings, and boronic acids also appear in drugs such as proteasome inhibitors and in sensors that bind sugars. Pinacol esters and MIDA boronates are more stable and easier to purify than free boronic acids, which is why they are often preferred for unstable heteroaryl partners.

Free boronic acids exist in equilibrium with their cyclic anhydrides (boroxines), so purity values can vary with drying and the method used. Ask how purity was measured, the water content and whether the material contains anhydride. Some heteroaryl boronic acids lose boron easily on storage (protodeboronation), so cold storage and fresh batches matter. For scale-up, ask about residual palladium and boron limits in downstream products.
