About fragment compounds

Fragment compounds are small, simple molecules used in fragment-based drug discovery. They usually follow the rule of three, meaning a molecular weight below 300, no more than three hydrogen bond donors and three acceptors, and a calculated logP of three or less. Fragment libraries contain hundreds to a few thousand such compounds chosen for diversity, solubility and synthetic tractability.

Because fragments are small, they bind weakly to protein targets, so screening uses sensitive biophysical methods such as X-ray crystallography, NMR, surface plasmon resonance and thermal shift assays. Hits are then grown or linked into potent leads. Newer libraries emphasise three-dimensional shape and include covalent fragments for reactive targets.

Ask for purity by LC-MS and NMR, solubility in water or buffer at the screening concentration, the format (dry powder, DMSO solution in plates or tubes) and the availability of follow-up analogues.
